Simplified RMID Model Building
Technické články | 2021 | MetrohmInstrumentace
Accurate and rapid material verification is critical in quality assurance and control across industries such as pharmaceuticals. Handheld Raman spectroscopy with chemometric models enables nondestructive analysis to confirm identity of active pharmaceutical ingredients and excipients. Streamlining model development increases accessibility for non-specialists and reduces time to deployment.
This work presents ModelExpert, an automated tool integrated into Mira Cal P software, designed to simplify and accelerate the construction of robust material verification models. The goal is to enable both expert and non-technical users to generate optimal chemometric parameters with minimal manual intervention.
The system employs the Metrohm Instant Raman Analyzer (MIRA P) Advanced, a handheld spectrometer featuring Orbital-Raster-Scan (ORS) technology and compliance with FDA 21 CFR Part 11. The Advanced Package includes attachments for direct sample analysis through containers and a vial holder for glass vials.
ModelExpert automates model building by iterating through 192 combinations of preprocessing steps (normalization, baseline correction) and principal component settings. It uses receiver operating characteristic (ROC) curves and the area under the curve (AUC) metric to rank parameter sets, penalizing combinations with undesirable false positive or negative rates.
ModelExpert presents optimized model parameters in a summary popup after evaluation. Users can generate a detailed PDF report containing ROC curves, validation statistics, and p-values. Applying the selected settings to training and validation datasets requires a single click, ensuring consistent and transparent model development.
ModelExpert within Mira Cal P streamlines chemometric model creation for handheld Raman spectroscopy, delivering robust, transparent, and efficient workflows. By automating parameter optimization and ROC-based validation, it democratizes advanced analytical techniques and accelerates material verification tasks across diverse industries.
